I have 2 questions?
1. Before sample preparation for mass spectrometry, is there any method that can be able to check sample for presence of protein or not?
2. Shotgun cloning differs from the clone-by-clone method in which of the following ways?
To check the presence of protein, we can use protein concentration assays for example Bradford assay or Lowry assay.
Clone by clone sequencing and clone by shotgun sequencing are two methods of modern-day genome sequencing. Clone by clone sequencing is a more reliable method of sequencing. It is a genomics analysis. For proteomics, shotgun proteomics is a use of bottom-up proteomics technique to study the whole proteins in a complex mixture. Protein mixtures are first digested by protease, and the resulting peptides are separated in HPLC, followed by tandem MS/MS analysis to identify the sequence of each peptide. The identified peptide sequences are compared with database, in searching for the corresponding protein identity. The name of shotgun proteomics is derived from shotgun sequencing of DNA
