1. The step for detection of oocyst in the midgut of the mosquito will be started from 1). Identify mosquito species 2). Kill the mosquito by putting them at -20 degrees Celcius, 3). move the killed mosquito to the glass slide, 4). 2 needles were used for mosquito dissection the first needle was used to fix the mosquito at the beginning of the abdomen and another one was used to pull at the 6 or 7 segments of the abdomen, 5). midgut was removed and used to detect oocyst under the microscope. Further, the sporozoite will detect in the salivary gland by 1). after the mosquito died, 2 needles were used for dissection, the first needle was used to fix the mosquito at the thorax and another one was used to pull at the head part, 2). get the salivary gland and put it on the glass slide and cover it with cover slip, 3). detection of sporozoite under the microscope.

Mosquito dissection
(+) Advantages: 1. highest specificity, 2. Suitable for the field examination of oocyst and sporozoite stage in the mosquitoes, 3. not expensive when compared with the other techniques.
(-) Disadvantages: 1. time-consuming for detection, 2. low sensitivity by low chance for getting infected mosquito in the field, 3. need expertise and well training for dissection.
Immunological assay
(+) Advantages: 1. high sensitivity and specificity for detection of an infected mosquito, 2. rapid and accurate detection.
(-) Disadvantages: 1. need laboratory equipment, 2. need technician, 3. more expensive, 4. not suitable for field examination.
Molecular technique
(+) Advantages: 1. more sensitivity and specificity for detection of an infected mosquito, 2. rapid and accurate detection, 3. can identify accurately Plasmodium species, 4. Just practice and no need for expertise.
(-) Disadvantages: 1. need laboratory equipment, 2. more expensive, 3. not suitable for field examination.
3. I will use the gold standard technique of “mosquito dissection” in the field examination because this technique did not needs much laboratory equipment and is not expensive. After getting the mosquito we can process them step by step in the field.
